冷冻和冻干血浆可用于局部凝血酶原时间系统校准、直接国际标准化比值(INR)测定和质量评估。本研究的目的是评估从接受维生素K拮抗剂治疗的患者获得的冷冻和冻干混合血浆中各种凝血活酶的INr的有效性。

研究人员使用各种凝血活酶和仪器，即凝血活酶国际标准以及从兔和牛脑制备的六种商业试剂和重组人组织因子，根据国际敏感性指数模型计算INRs。使用凝血时间和国际标准工业指数值的标准偏差来评估INRs的不确定性。根据批准的临床和实验室标准协会(CLSI)指南EP30-A评估血浆的可交换性。根据国际血栓形成和止血学会(SSC/ISTH)科学和标准化委员会的抗凝控制小组委员会指南，对用于直接INR测定的六个冷冻血浆混样进行验证。

研究结果显示，对于所有冷冻和冻干血浆，用牛凝血活酶试验计算的INR低于用其他凝血活酶计算的INR。除了少数例外，冷冻和冷冻干燥的混合血浆是可交换的。当六个冷冻血浆混样用于局部校准时，除血栓测试外，所有商业试剂的INR分析偏差小于10%。

研究结果表明，处理新鲜血浆制备混合冷冻血浆和冷冻干燥血浆可能导致不同的INR分配，这取决于所用的凝血活酶。尽管INR略有差异，但一套六个冷冻血浆混样可用于通过直接INR测定进行局部校准。
